Quaternary Ammonium Palmitoyl Glycol Chitosan (GCPQ) Loaded with Platinum-Based Anticancer Agents—A Novel Polymer Formulation for Anticancer Therapy

چکیده
Quaternary ammonium palmitoyl glycol chitosan (GCPQ) has already shown beneficial drug delivery properties and been studied as a carrier for anticancer agents. Consequently, we synthesised cytotoxic platinum(IV) conjugates of cisplatin, carboplatin oxaliplatin by coupling via amide bonds to five GCPQ polymers differing in their degree palmitoylation quaternisation. The were characterised 1H 195Pt NMR spectroscopy well inductively coupled plasma mass spectrometry (ICP-MS), the latter determine amount units per polymer. Cytotoxicity was evaluated MTT assay three human cancer cell lines (A549, non-small-cell lung carcinoma; CH1/PA-1, ovarian teratocarcinoma; SW480, colon adenocarcinoma). All displayed high increase activity factors up 286 times compared corresponding complexes mostly outperformed respective platinum(II) counterparts 20 times, also taking into account loading Finally, biodistribution experiment performed with an oxaliplatin-based conjugate non-tumour-bearing BALB/c mice revealing increased accumulation tissue. These findings open promising opportunities further tumouricidal studies especially focusing on

Polymeric biomaterials for the delivery of platinum-based anticancer drugs.
Since cisplatin, cis-diamminedichloroplatinum(ii), received FDA approval for use in cancer treatment in 1978, platinum-based drugs have been one of the most widely used drugs for the treatment of tumors in testicles, ovaries, head and neck. However, there are concerns associated with the use of platinum-based anticancer drugs, owing to severe side effects and drug resistance. In order to overco...
متن کاملNetwork-based approaches for anticancer therapy (Review).
Cancer is a complex disease resulting from alterations of multiple signaling networks. Cancer networks have been identified as scale-free networks and may contain a functionally important key player called a hub that is linked to a large number of interactors. Since a hub can serve as a biological marker in a given network, targeting the hub could be an effective strategy for enhancing the effi...

Platinum-based anticancer drugs are one of the most widely used drug classes in cancer therapy. Almost half of the chemotherapy regimens used today contains a platinum drug, such as cisplatin or carboplatin. However, the drug resistance and non-specific cytotoxicity ultimately limits broader application of these drugs. Improvement of drug targeting and delivery systems are effective approaches ...
متن کاملpH-responsive glycol chitosan-cross-linked carboxymethyl-β-cyclodextrin nanoparticles for controlled release of anticancer drugs.
Carboxymethyl-β-cyclodextrin (CMβ-CD)-modified glycol chitosan (GCS) nanoparticles (GCS-CMβ-CD NPs) were synthesized, and their pH-sensitive drug-release properties were investigated. GCS-CMβ-CD NPs could encapsulate doxorubicin hydrochloride (DOX), and the encapsulation efficiency and loading capacity increased with the amount of CMβ-CD. Drug-release studies indicate that DOX released was grea...

In the 1920s, when the genetic theory of cancer was taking root, German biochemist Otto Warburg (winner of the 1931 Nobel prize in physiology or medicine) proposed that abnormal energy metabolism caused cancer. Cells can generate energy either through oxidative respiration in mitochondria or through glycolysis in the cytoplasm. Warburg showed that tumors have an acidic extracellular environment...
